چکیده
Tissue necrosis may be detected by the appearance of intracellular enzymes in extracellular spaces (Kibe & Nilsson, 1967) and this principle has been applied to ischaemic and hypoxic cardiac necrosis in man and animals (Roe & Starmer, 1975; Shell et al., 1973). Beating myocardial cells in culture represent an extremely reproducible preparation for studies of the deleterious effect of hypoxia on cardiac tissue. The aim of the study reported here was to examine the metabolic basis of the exacerbation of damage caused by increased medium CaZf concentration to heart cells subjected to anoxia.

Antifibrinolytic Activities of or-iV-Acetyl-L-Lysine Methyl Ester, e-Aminocaproic Acid, and Tranexamic Acid Importance of Kringle Interactions and Active Site Inhibition
cr-jV-acetyl-L-lysine methyl ester (NALME) is a lysine analogue that reportedly binds to low-affinity lysine binding sites in plasmin(ogen) and miniplasmin(ogen). In the studies presented here, we show that NALME has antifibrinolytic activity; however, unlike the therapeutic agents e-amino-n-caproic acid (eACA) and tranexamic acid (TEA), the activity of NALME is based on inhibition of the plasm...
متن کاملStreptococcal Fibrinolysis: a Proteolytic Reaction Due to a Serum Enzyme Activated by Streptococcal Fibrinolysin
1. Methods for the preparation and partial purification of streptococcal fibrinolysin are described. 2. The lysis of fibrin clots in the presence of streptococcal fibrinolysin is associated with proteolysis of the fibrin. Digestion is due to an enzyme normally present in serum or plasma in an inactive state, which is activated by fibrinolysin. Fibrinolysin alone has no demonstrable proteolytic ...
متن کاملIn vivo fibrinolytic activity and pharmacology of various plasmin (fibrinolysin) preparations.
A quantitative method based on the disappearance of I laljeled fibrin h;is been used to study the in vivo effect of fibrinolytic agents. Various preparations of human and bovine plasmin were found to lyse clots effectively. The minimal effective dose of human plasmin prepared by the method of Kline was 15 Loomis units/Kg. Plasmin preparations given intravenously produced hypotension and leukope...
متن کاملBiochemical Studies on the Fibrinolytic Activity of Hemolytic Streptococci
The fibrinolysin of hemolytic streptococci exerts no hydrolytic action upon casein, gelatin, or peptone. The action on solid human fibrin is characterized by a small and gradual increase in the amino nitrogen content of the solution. The specific and special enzymatic action of fibrinolysin is contrasted with trypsin and with streptococcal peptase (4). Solutions of human fibrinogen, after brief...
متن کاملEarly events in the plasmin digestion of fibrinogen and fibrin. Effects of plasmin on fibrin polymerization.
We have studied the effects of limited plasmic digestion of fibrinogen and fibrin on the physical properties of fibrin gels. After limited digestion the rigidity (elasticity) of the clot formed from degraded fibrinogen is 1% of the rigidity obtained from undegraded fibrinogen, whereas the decrease in clottability as a result of this limited digestion is only about 10%. When plasmin digests fibr...
